Tissue sections can contain endogenous biotin, biotin binding proteins, lectins or non-specific binding substances. Tissues may also bind avidin, biotin, peroxidase biotin, peroxidase streptavidin in avidin biotin based Immunohistochemistry. This endogenous biotin and avidin will give high background in the absence of biotinylated secondary antibodies. It may be necessary to block the tissue with avidin, followed by biotin. These reagents are added before the primary antibody.

  • One 15 ml bottle of diluted avidin in a phosphate buffered saline solution.

  • One 15 ml bottle of diluted biotin in a phosphate buffered saline solution.

  • For  In Vitro diagnostic use.

  • This product is intended for the blocking endogenous biotin found in tissue used in immunohistochemistry, immunofluorescence, and similar mammalian tissue based techniques. Some tissue sections contain endogenous biotin, biotin binding proteins, lectins or non-specific binding substances. Tissues may also bind avidin, biotin, peroxidase biotin, peroxidase streptavidin in avidin biotin based Immunohistochemistry. These tissues will give high background in the absence of biotinylated secondary antibodies. It may be necessary to block the tissue with avidin, followed by biotin. These reagents are added before the primary antibody.

  • Directions for use

  •  After incubating in normal serum, directly prior to incubating in primary antibody, wipe off back and sides of slide with a kimwipe, lay flat on benchtop (in a humidity chamber if desired) and cover tissue with 1-3 drops of avidin block.  Incubate RT 15 min., checking every 5 min. to make sure samples don’t dry out. If starting to dry, add more avidin block.  Continue by washing slide 1X 3 min in wash buffer, then incubate in biotin block for 15 min at RT, making sure drying does not occur. Wash in wash buffer 3 X 3 min at RT. Incubate in primary antibody.

  • Store at 4°C. Do not freeze. Discard unused reagent after expiration.

  • Precautions

  • For professional users.

    1. Follow storage conditions as to minimize the possibility of microbial contamination of reagents.
    2. As with any product, proper handling procedures should   be used.
    3. Wear appropriate protective equipment to avoid contact with eyes and skin.

  • Notes

  • This product does not contain preservatives. Preservatives such as sodium azide inhibit key enzymes involved in IHC technology such as peroxidases. This quenching can cause the occurrence of false negative results. Due to the absence of preservatives, it is advised to keep strict adherence to storage conditions, proper handling, and disposal of the reagent after expiration occurs.
