
| At Prohisto, we noticed a recurring problem of IHC: often, mounted tissues stained unevenly, giving inconsistent results, and reducing our confidence. We were never sure if a lack of positive cells were the true result, or if negative areas on slides were from shortcomings of the traditional "dropping on" technique. We decided that the best method of antibody-antigen recognition was already in use every day, in every part of the world-in WESTERN BLOTTING. In that tried and true method, the membrane, with the bound antigen , is submerged in an antibody solution. Prohisto's patent pending apparatus, the Antibody Amplifier, uses a SUBMERSION method to SATURATE the mounted tissue with antibody. The antibody is diluted in milliliters of buffer solution instead of microliters. Although this sounds more expensive, it is not, because the antibody can be diluted up to 30,000 times the manufacturer's recommended dilution! We have also shown the diluted antibody can be frozen and reused several times, similar to western blotting, with no loss of efficacy. Your slides are also rocked on a standard rocker or orbital mixer, guaranteeing that the antibody has ample opportunity to reach its target epitope. When you have finished your protocol, and are viewing results, you can have confidence that the method has not interfered with results. Prohisto takes pride in improving immunohistochemistry.
