The Antibody Amplifier™ improves IHC in 4 ways:
1.) Increases Antibody Dilution
2.) Promotes Even Staining
3.) Enhances Reproducibility
4.) Freeze and Re-use Antibody Multiple Times
INCREASES ANTIBODY DILUTION
ENHANCED EVEN STAINING
INCREASED REPRODUCIBILITY
These pictures depict a comparison of the "standard" i.e. "dropping
on" method versus the PROHisto method using the Antibody Amplifier.
REUSE YOUR ANTIBODY
Figure. Shows the improved efficiency with the Antibody AmplifierTM
using less concentrated antibody. The antibody here was
anti-phospho-pRb Ser 780 (polyclonal, Cell Signaling). These are
paraffin-embedded, serial sections from mouse colon.
Figure. Shows the improved quality of staining and even staining
patterns using the Antibody AmplifierTM. On the left is the standard
sedentary method, where there is uneven staining. The right side of the
picture shows that using the Antibody AmplifierTM results in even and
reproducible staining. We used the anti-phospho-pRb Ser 780
(polyclonal, Cell Signaling). These are paraffin-embedded, serial
sections from mouse colon. For the STANDARD method (left), the
antibody concentration was 1:1000. For the Antibody AmplifierTM
method (right) (rocking the slides), the antibody concentration was
1:100,000.

Figure. Shows increased consistency and improved reproducibility of
staining with the Antibody AmplifierTM. These are serial sections. On the
left, arrows indicate where there are clear differences in staining patterns in
serial sections. Alternatively, the right side shows the consistent staining
pattern using the Antibody AmplifierTM. We used the anti-phospho-pRb Ser
780 (polyclonal, Cell Signaling). These are paraffin-embedded, serial
sections from mouse colon. For the STANDARD method (left), the antibody
concentration was 1:1000. For the Antibody AmplifierTM method (right)
(rocking the slides), the antibody concentration was 1:100,000.
Figure. Using the Antibody AmplifierTM, you can re-use
antibodies up to 4 times without loosing efficiency. We used
the anti-phospho-pRb Ser 780 (polyclonal, Cell Signaling,
1:100,000). These are paraffin-embedded, serial sections
from mouse colon.
